Abstract In this study, we focused on the key hypoglycemic oligosaccharide components derived from thinned peach crude polysaccharides (CPPS), elucidated their structural characteristics and hypoglycemic mechanisms, and improved the utilization of this agricultural waste. CPPS was fractionated via anion-exchange chromatography and ultrafiltration, yielding 2 key components (W1-2 and W2-2) with enhanced α -glucosidase inhibition and half maximal inhibitory concentration (IC 50 ) were 0.57 and 0.37 mg/mL, respectively. They improved glucose consumption, glycogen synthesis, hexokinase, and pyruvate kinase activities in insulin resistant HepG2 cells. Structural analysis using LC-MS and methylation revealed W1-2 contains disaccharides where galactose or glucose is linked to terminal arabinose through 1,4-glycosidic bonds, alongside arabinose-rich pentasaccharides featuring T-Ara f , 1,5-Ara f and 1,3,5-Ara f residues. W2-2 comprises trehalose-like disaccharides with accounting for 76.95% of T-Glc p and arabinose pentasaccharides with similar branching patterns. These findings suggest that trehalose-like disaccharides and arabinose-derived pentasaccharides in CPPS may play a key role in blood glucose regulation. This study offers important insights into the roles of bioactive carbohydrates in diabetes intervention.

Abstract In this study, antioxidant peptides were isolated and purified from Tongchuan Douchi through ultrafiltration and gel chromatography. To further assess the in vivo antioxidant activity of the fraction (F3) which had a high DPPH radical scavenging rate, 60 male mice were randomly divided into 6 groups (normal, positive control, model group, and low-, medium-, and high-dose group), and immunosuppression was induced via intraperitoneal cyclophosphamide injection. Hepatorenal function was evaluated by determining organ coefficients and oxidative stress markers including total superoxide dismutase (T-SOD), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) levels. The results showed that 10 peptides (LLLPH, KDFPPR, DDFFL, ADFYNPK, IIGGI, DTPMIG, SVVDM, LVSPP, C(+57.02)ADLF, and VIVEL) which are rich in hydrophobic amino acids were identified in the fraction F3 by LC-MS/MS. The quantitative analysis of the animal experiments demonstrated that administration of medium and high doses of the F3 fraction significantly ( P < 0.05) improved the liver, kidney, and spleen coefficients in immunosuppressed mice compared to the model group. The high-dose F3 administration significantly increased the SOD and GSH-Px activities while reducing the MDA levels in both hepatic and renal tissues of mice. These results revealed that Douchi-derived peptides can mediate antioxidant effects in vivo by both scavenging free radicals and activating endogenous antioxidant enzymes.

Lilium polysaccharides (LLP) exhibited various biological activities. However, the low bioavailability of polysaccharides limits their effectiveness in practical applications. To enhance their functionality and bioavailability, polysaccharides from Lanzhou lily (Lilium davidii var. willmottiae (E.H. Wilson) Raffill) bulbs were purified and chemically synthesized into polysaccharide-Fe (III) complexes in this study. Then, the structural characterization, physicochemical properties, thermal stability and digestibility of LLP-Fe (III) were systematically evaluated to assess its potential as an iron supplement. Results showed that LLP-Fe (III) had a higher molecular weight than LLP, with a monosaccharide composition dominated by mannose and glucose at a molar ratio of 1:1.23. LLP-Fe (III) was also revealed to have significantly enhanced thermal stability. FT-IR analysis confirmed the chelation between Fe (III) and carboxyl and hydroxyl groups in LLP through stable Fe−OOH bonds. Scanning electron microscopy displayed a dense morphology with smooth and small particles in LLP-Fe (III). Furthermore, in vivo simulated digestion tests indicated superior gastrointestinal stability and acid tolerance of LLP-Fe (III). These findings suggested that the polysaccharide-Fe (III) complexes hold promise as a novel iron supplement with potential health benefits.

Dried shrimp odour deterioration is closely linked to microbial metabolism in various body parts, especially the cephalodorax viscera. However, the microorganisms responsible for this process and the key off-flavor compounds (KOFCs) remain poorly understood. This study examined the microbial and flavor characteristics of head (DH), meat (DM), and whole shrimp (DW). Bacillus (38.17%) and Taeseokella (12.14%) predominated in DW and DH at the initial stage, respectively, but were replaced later by Photobacterium (DW: 18.14%, DH: 10.15%) and Vibrio (DW: 6.87%, DH: 5.68%). As microbiota dynamics evolved, the types and concentrations of volatile compounds increased in both DW and DH but not in DM, which was dominated by Staphylococcus at all stages. A totaly of 39 KOFCs were found to differentiate the 3 groups, among which ammonium carbamate was first identified in dried shrimp. Further correlation analysis showed that Photobacterium and Vibrio in the cephalodorax viscera were the primary microorganisms causing flavor deterioration. This work offers novel perspectives on the mechanisms underlying shrimp deterioration.

Red kidney beans (Phaseolus vulgaris L.), a nutrient-dense legume, face significant limitations in food applications due to persistent allergenicity. The dual effects of industrial autoclaving (121 °C, 15 min) on allergenicity modulation were evaluated through in vitro gastrointestinal digestion models in this study, integrated with sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), immunoblotting, and computational epitope prediction. Electrophoretic profiling demonstrated partial degradation of major allergens for 43 kDa phaseollin-like protein, lectin subunits (31/25 kDa). Legumin α-subunits exhibited pepsin resistance (74.8% stability), contrasting with rapid degradation of α-amylase inhibitor (α-AI) and group 3 late embryogenesis abundance (G3LEA) proteins. Chromatographic immunoreactivity screening revealed autoclaved samples generated 8 antigenic fractions, including novel hydrophobic peptides with enhanced immunoglobulin E (IgE)-binding potential. Mass spectrometry identified 849 and 1 516 unique peptides in raw and autoclaved digests, respectively, with phaseollin and lectins dominating post-processing epitopes. Bioinformatics highlighted conserved allergenic motifs (e.g., VLVKPIQIR, LPQQADAE) in phaseollin and lectins, alongside cross-reactive sequences homologous. Paradoxically, autoclaving reduced intact allergen content but amplified immunoreactive diversity through conformational exposure of epitopes and hydrophobic residue enrichment. These findings underscore the necessity for epitope-specific risk assessment in legume processing, balancing benefits of thermal intervention against unintended allergenic consequences.
